چکیده

Natalizumab, a humanized monoclonal antibody against very late antigen-4 (VLA-4), inhibits the transmigration of immune cells across the bloodbrain barrier into the central nervous system (CNS). However, progressive multifocal leukoencephalopathy (PML) is caused following natalizumab treatment in patients with relapsing remitting multiple sclerosis (RRMS). The reason for higher PML prevalence in patients treated with natalizumab in Europe than those in the USA patients may not be the more frequent use of cytotoxic medications in Europe, but may be the lower body weight (BW) of European patients and higher nadir serum concentrations of natalizumab. We recommended BW-based natalizumab therapy in RRMS patients because a linear relation exists between serum concentrations and BW (50 110 kg) (y1⁄4 0.5363xþ 75.71, R1⁄4 0.846). Recently, extended interval dosing of natalizumab therapy has been shown.

Correlation of Midkine Serum Level with Pro- and Anti-Inflamatory Cytokines in Multiple Sclerosis

Background: Midkine (MK) is a heparin-binding growth factor with promoting effects in inflammatory responses through enhancing leukocytes migration. Objective: To study the correlation between MK serum levels and concentration of inflammatory cytokines in Multiple Sclerosis (MS) patients. Methods: We evaluated the MK level and its relationship with inflammatory cytokines (IL-17 and IL-23) and a...

Natalizumab treatment decreases serum IgM and IgG levels in multiple sclerosis patients.

BACKGROUND Treatment with natalizumab, a humanized monoclonal antibody against alpha4beta1 integrin, is associated with an increase in lymphoid progenitor cells and B cells in peripheral blood. OBJECTIVE The objective of this study was to examine the impact of natalizumab therapy on serum levels of total IgG, IgA and IgM in patients with multiple sclerosis (MS). METHODS In two independent c...

Clinical relevance of serum natalizumab concentration and anti-natalizumab antibodies in multiple sclerosis.

BACKGROUND Antibodies against natalizumab have been found in 4.5-14.1% of natalizumab-treated multiple sclerosis (MS) patients. If antibodies persist, they are associated with an adverse effect on treatment response. However, it has proved to be difficult to standardize anti-drug antibody measurements. OBJECTIVES The purpose of this study was to evaluate the clinical and radiological impact o...

CD4 cell response to interval therapy with natalizumab

Natalizumab treatment alters peripheral CD4 cells counts in multiple sclerosis (MS) patients, providing a way to monitor the pharmacodynamic effects of the drug. The study was undertaken to assess whether CD4 cell counts correlate with different phases of natalizumab treatment of relapsing MS patients, including during a 12-week planned treatment interruption, and whether that might provide ins...
